In questa lettera, che immagino sia stata consegnata dalla residenza del Granduca come da ultima frase, c’è a "Magistrato Supremo" allegato all’introduzione del Granduca. Immagino che questo ufficio servisse ancora allo stesso scopo dei tempi di Cosimo I de’ Medici, ovvero il più alto organo dell’amministrazione statale del Granducato?
Mi chiedo se "Sotto-Cancelliere" qui c’era una specie di posto di segreteria? La Wikipedia italiana ha identificato Francesco Seratti come a "primo ministro" del Granducato fino al 1798 con l’incarico di "Segretario di Stato degli Affari Interni e Stranieri" – So che alcuni paesi hanno cancellieri a capo del governo. Tuttavia, sarebbe un po’ bizzarro mettere il file "Sotto-Cancelliere" come a "vicepremier" in questo caso?

“Sigg.” is short for “Signori” (in Italian, when an abbreviation is pluralized, you double the last letter. So “Sig.” for “signore”, “sigg.” for “signori”), meaning literally “Misters” but generally used as a sign of respect. You could also potentially interpret it as “lord”, since that’s also “signore”.
So “SIGG. L. T. E CONSIGLIERI” means “Mister L. T. and the Councilors”.
I’m not sure if “L. T.” is a name or a rank. Could stand for “luogotenente”, meaning “lieutenant”.
The full heading could be translated as “to [the attention of] the most illustrious and most fair, the lords L.T. and the councillors”
